Each microbial strain has the potential to produce multiple compounds, but only subsets of these compounds are made under specific growth conditions. Therefore, variations in cultivation parameters can elicit the production and discovery of new secondary metabolites by changing cultivation parameters such as media composition, various nutrients, trace elements, physical parameters (i.e., pH, temperature), and chemical elicitors (i.e., sub-lethal concentrations of antibiotics, communication molecules). Moreover, the co-cultivation of microbes and the addition of factors affecting epigenetic control can also be framed within the OSMAC principle.

| Experiment Detail |
Branches of Picea mariana and Picea rubens of various age classes were collected in New Brunswick, Canada. A 2 % MEA slant of Diaporthe maritima DAOM 628553 was macerated in sterile distilled deionized water (ddH2O) and an aliquot was used to inoculate (5 % v/v) 15 × 250 mL Erlenmeyer flasks containing 50 mL of 2 % ME (Bacto, Becton, Dickinson and Company, Le Pont de Claix, France) broth. Flasks were incubated on a rotary shaker (100 rpm) for 1 wk in the dark at 25 °C. Cultures were macerated, individually transferred to Glaxo bottles containing 1 L of the same liquid medium, and incubated without agitation as described above for 7 wk.

Fungal culture: The strain D. terebinthifolii LGMF907 was obtained from leaves of S. terebinthifolius Raddi collected in Curitiba, Parana, Brazil (25° 28′ 31.847″ S, 49° 17′ 5.629″ W). Strain LGMF907 was grown on potato dextrose agar (PDA; IBI Scientific) plates at 28 ℃ for 15-20 days. Large-scale fermentation: The large-scale cultivation was performed by inoculation of 24 agar plugs into 2-L Erlenmeyer flasks containing 800 mL of malt extract broth (MEB; 20 g/L malt extract, 20 g/L glucose, and 1 g/L peptone), with a total of 20 L of culture. The flasks were incubated for 10 days, at pH 5.8, 28 ℃, and 120 rpm.

Specimen examined: Brazil, Corumba, Pantanal, Mato Grosso do Sul, S19° 32′ 36.9″ , W57° 02′ 21.8″, endophytic species isolated from leaf of Vochysia divergens (popular name Cambara), 13 June 2016. Strain LGMF1583 cultivation: Strain LGMF1583 was cultivated in PDA at 28 ℃ for 3 days, and culture was cut into 8 mm plugs. Five plugs were used to inoculate 32 Erlenmeyer flasks (500 mL) containing 250 mL MEB medium and incubated at 28 ℃ for 10 days on a rotary shaker (180 rpm).
